Fluorescence-activated cell sorting (FACS) is a specialized type of flow cytometry. It provides a method for sorting a heterogeneous mixture of biological cells into two or more containers, one cell at a time, based upon the specific light scattering and fluorescent characteristics of each cell. It is a useful scientific instrument as it provides fast, objective and quantitative recording of fluorescent signals from individual cells as well as physical separation of cells of particular interest. The acronym FACS is trademarked and owned by Becton, Dickinson and Company. Among the large majority of researchers who use this technology for sorting or analysis, this term has become generic in common usage, much like xerox or kleenex. The first cell sorter was invented by Mack Fulwyler in 1965, using the Coulter principle, a relatively difficult technique that is no longer used in modern instruments. The technique was expanded by Len Herzenberg, who was responsible for coining the term FACS. Herzenberg won the Kyoto Prize in 2006 for his seminal work in flow cytometry. The cell suspension is entrained in the center of a narrow, rapidly flowing stream of liquid. The flow is arranged so that there is a large separation between cells relative to their diameter. A vibrating mechanism causes the stream of cells to break into individual droplets. The system is adjusted so that there is a low probability of more than one cell per droplet. Just before the stream breaks into droplets, the flow passes through a fluorescence measuring station where the fluorescent character of interest of each cell is measured. An electrical charging ring is placed just at the point where the stream breaks into droplets. A charge is placed on the ring based on the immediately prior fluorescence intensity measurement, and the opposite charge is trapped on the droplet as it breaks from the stream. The charged droplets then fall through an electrostatic deflection system that diverts droplets into containers based upon their charge. In some systems, the charge is applied directly to the stream, and the droplet breaking off retains charge of the same sign as the stream. The stream is then returned to neutral after the droplet breaks off.

When it was all said and done, we put in about 2.5 hours!! Questions were coming in left and right from the chat room so the videos will seem long for folks who weren't there in person. To quickly explain what was going on, I planned on a quick shop tour and a little Q&A. Nicole was operating the camera and Mom was moderating the chat room and yelling out questions as they came in. It was pretty funny. Everything went well, minus a few technical issues. Unfortunately, the live audio was via my laptop microphone. I have it all figured out at this point so the audio will be much better next time. As you will see though, the recording came out as expected. ExoTEST is a patented double sandwich ELISA assay for quantitative and qualitative analysis of exosomes. In particular ExoTEST is a successful platform for exosomes quantification and characterization from small amount of human biological fluids or cell media and it may be exploited to identify exosomes released by cancer cells in the plasma and urine of tumor patients in various disease conditions. Google Workshop on Quantum Biology A Quantum of Solace: Molecular Electronics of Benzodiazepines Presented by Luca Turin October 22, 2010 ABSTRACT Benzodiazepines and related drugs modulate the activity of GABA-A receptors, the main inhibitory receptor of the central nervous system. The prevailing view is that these drugs bind at the interface between two receptor subunits and allosterically modulate the response to GABA. In this talk I shall present evidence that benzodiazepines work instead by facilitating electron transport from the cytoplasm to a crucial redox-sensitive group in the gamma subunit. If this idea is correct, benzodiazepines should not only be regarded as keys fitting into a lock, but also as one-electron chemical field-effect transistors fitting into an electronic circuit. During the lecture, Dr. Zulueta-Fülscher introduced A Practical Guide to Constitution Building (2011) as well as the ConstitutionNet website where users can find all knowledge resources produced by International IDEA dealing with design-related issues such as federalism, bicameralism, constitutional assessment, constitutional reform processes and the role of political parties, among many others. This public lecture was organized by the Department of Political Science and the Institute of Government and Law Reform, UP Law Center and co-sponsored by the Third World Studies Center.
